Succinate, an intermediate metabolite of the Krebs cycle, can alter metabolomics response to certain drugs and controls array molecular responses in urothelium through activation its receptor, G-protein coupled receptor 91 (GPR91). Mirabegron, a β3-adrenergic (β3-AR) agonist used treat overactive bladder syndrome (OAB), increases intracellular cAMP detrusor smooth muscle cells (SMC), leading relaxation. We have previously shown that succinate inhibits forskolin-stimulated production urothelium. To determine whether interferes with mirabegron-mediated relaxation, we examined their individual synergistic effect urothelial-cell SMC signaling. first confirmed β3-AR involvement mirabegron by quantifying abundance immunoblotting cultured urothelial cellular localization immunohistochemistry rat tissue. Mirabegron increased levels but not cells, increase was inhibited succinate, suggesting it impairs cAMP-mediated relaxation mirabegron. Succinate inducible nitric oxide synthesis secretion only release indirectly induces exposure decreased expression protein whole vivo vitro, indicating this may lead impaired pharmacodynamics bladder. Together, our results demonstrate settings metabolic stress (e.g., syndrome) interaction, inhibition production, ultimately requiring dose adjustment for treatment OAB related these conditions.
